Steve Dolan's side beat Hawick Royal Albert 3-1 last Saturday.
Robert Scott put the home side ahead in the first half only for a Lewis Dunbar own goal to level things up before the break.
Dunbar put Vale ahead again midway through the second half and Scott's second of the match sealed the win with 12 minutes remaining.
Glenafton will visit Princess Royal Park on Saturday, October 14.
